Aptamers are unique short DNA or RNA oligomers with a broad variety of potential applications in molecular biology and biotechnology, particularly as tools to study cellular and subcellular events. In this paper, we describe the preparation and characterization of a library of 9-deazadenosine- or 8-azido-9-deazadenosine-containing DNA sequences, which, after separation by reverse-phase HPLC and 5'-excision-coupled RNase H, were used to select for novel aptamers. We found that the pool of aptamers selected from 8-azido-9-deazadenosine-containing DNA sequences preferentially bound to the nuclear RNA-binding protein, FUS. After initial selection, we show that the aptamers can be expanded by a stepwise process of selection and amplification. The biological functions of the aptamers were assessed by their ability to modulate the transcription of two genes, human growth hormone and c-fos. A single aptamer was able to simultaneously induce increased transcription of these two genes. We also show that a variety of other known aptamers with unrelated secondary structures can be expanded in this manner.Q:
